    Synthetic lethal analysis of Caenorhabditis elegans posterior embryonic patterning genes identifies conserved genetic interactions

    Phenotypic robustness is evidenced when single-gene mutations do not result in an obvious phenotype. It has been suggested that such phenotypic stability results from 'buffering' activities of homologous genes as well as non-homologous genes acting in parallel pathways. One approach to characterizing mechanisms of phenotypic robustness is to identify genetic interactions, specifically, double mutants where buffering is compromised. To identify interactions among genes implicated in posterior patterning of the Caenorhabditis elegans embryo, we measured synthetic lethality following RNA interference of 22 genes in 15 mutant strains. A pair of homologous T-box transcription factors (tbx-8 and tbx-9) is found to interact in both C. elegans and C. briggsae, indicating that their compensatory function is conserved. Furthermore, a muscle module is defined by transitive interactions between the MyoD homolog hlh-1, another basic helix-loop-helix transcription factor, hnd-1, and the MADS-box transcription factor unc-120. Genetic interactions within a homologous set of genes involved in vertebrate myogenesis indicate broad conservation of the muscle module and suggest that other genetic modules identified in C. elegans will be conserved

    Maternal and zygotic activities of the homeodomain protein PAL-1 specify the identity and maintain the development of the multipotent C blastomere lineage in the C. elegans embryo. To identify PAL-1 regulatory target genes, we used microarrays to compare transcript abundance in wild-type embryos with mutant embryos lacking a C blastomere and to mutant embryos with extra C blastomeres. pal-1-dependent C-lineage expression was verified for select candidate target genes by reporter gene analysis, though many of the target genes are expressed in additional lineages as well. The set of validated target genes includes 12 transcription factors, an uncharacterized wingless ligand and five uncharacterized genes. Phenotypic analysis demonstrates that the identified PAL-1 target genes affect specification, differentiation and morphogenesis of C-lineage cells. In particular, we show that cell fate-specific genes (or tissue identity genes) and a posterior HOX gene are activated in lineage-specific fashion. Transcription of targets is initiated in four temporal phases, which together with their spatial expression patterns leads to a model of the regulatory network specified by PAL-1

    A UV to Mid-IR Study of AGN Selection

    We classify the spectral energy distributions (SEDs) of 431,038 sources in the 9 sq. deg Bootes field of the NOAO Deep Wide-Field Survey (NDWFS). There are up to 17 bands of data available per source, including ultraviolet (GALEX), optical (NDWFS), near-IR (NEWFIRM), and mid-infrared (IRAC/MIPS) data, as well as spectroscopic redshifts for ~20,000 objects, primarily from the AGN and Galaxy Evolution Survey (AGES). We fit galaxy, AGN, stellar, and brown dwarf templates to the observed SEDs, which yield spectral classes for the Galactic sources and photometric redshifts and galaxy/AGN luminosities for the extragalactic sources. The photometric redshift precision of the galaxy and AGN samples are sigma/(1+z)=0.040 and sigma/(1+z)=0.169, respectively, with the worst 5% outliers excluded. Based on the reduced chi-squared of the SED fit for each SED model, we are able to distinguish between Galactic and extragalactic sources for sources brighter than I=23.5. We compare the SED fits for a galaxy-only model and a galaxy+AGN model. Using known X-ray and spectroscopic AGN samples, we confirm that SED fitting can be successfully used as a method to identify large populations of AGN, including spatially resolved AGN with significant contributions from the host galaxy and objects with the emission line ratios of "composite" spectra. We also use our results to compare to the X-ray, mid-IR, optical color and emission line ratio selection techniques. For an F-ratio threshold of F>10 we find 16,266 AGN candidates brighter than I=23.5 and a surface density of ~1900 AGN per deg^2.Comment: Submitted to ApJ, 35 pages, 17 figures, 2 table

    Quantum Cosmology of Kantowski-Sachs like Models

    The Wheeler-DeWitt equation for a class of Kantowski-Sachs like models is completely solved. The generalized models include the Kantowski-Sachs model with cosmological constant and pressureless dust. Likewise contained is a joined model which consists of a Kantowski-Sachs cylinder inserted between two FRW half--spheres. The (second order) WKB approximation is exact for the wave functions of the complete set and this facilitates the product structure of the wave function for the joined model. In spite of the product structure the wave function can not be interpreted as admitting no correlations between the different regions. This problem is due to the joining procedure and may therefore be present for all joined models. Finally, the {s}ymmetric {i}nitial {c}ondition (SIC) for the wave function is analyzed and compared with the ``no bouindary'' condition.     Fear of the unknown: a pre-departure qualitative study of Turkish international students

    This paper presents findings from eleven in-depth interviews with Turkish undergraduate students, who were, by the time of data collection, about to spend a semester at a European university under the Erasmus exchange scheme. The students all agreed to be interviewed about their feelings about studying in a foreign culture, and were found to be anxious prior to departure about the quality of accommodation in the new destination, their language ability and the opportunity to form friendships. Fears were expressed about possible misconceptions over Turkey as a Muslim and a developing country. Suggestions are made for HEI interventions to allay student travellers’ concerns

    International education: a force for peace and cross-cultural understanding?

    This paper discusses the notion that the international sojourn has the potential to transform sojourners into cultural mediators who carry the power to improve global relations. A year-long ethnographic study of the adjustment experiences of international postgraduate students in England revealed a universal early enthusiasm for cross-cultural contact that was matched by a widespread adoption of segregated patterns of interacting. The most common friendship networks were described by bonds with conationals, and yet all students attested to an increase in their cultural learning and mindfulness by the end of the sojourn. Nevertheless, intercultural competence was maximised only in those few students who pursued a multicultural strategy of interaction, leading the researcher to call on Higher Education Institutions to instigate policies to encourage lasting cross-cultural contact

    Breast Metastasis from Differentiated Thyroid Carcinoma Primary: A Case Report and Review of Literature

    Introduction: Metastasis to the breast from non-mammary malignancy is rare, with a reported incidence of less than 1%[2-4]. Aside from hematologic malignancies and contralateral breast cancer, the most common primary malignancies that metastasize to the breast are lung, ovarian, gastrointestinal cancers, and melanoma.Presentation of Case: We present an 82-year-old female with metastatic breast cancer arising from a primary papillary thyroid carcinoma. She presented with a palpable right breast mass 4 years after resection and treatment of a primary thyroid cancer.Conclusion: Ultrasound guided core biopsy and immunohistochemical staining demonstrated metastatic papillary thyroid carcinoma. The patient underwent lymphoscintigraphy and subsequent resection of the breast mass along with two axillary lymph nodes

    The First Impressions program was designed to help communities learn about their strengths and shortcomings through the fresh eyes of first-time visitors. This Extension-led community assessment program has been implemented in over 500 communities since the 1990s, yet scant literature exists regarding the program\u27s theoretical foundations or best practices for implementation or evaluation. To address increasing interest in the program among Extension professionals nationwide, we aim to encourage new scholarship regarding the program\u27s use and impacts by providing an overview of how the program works; a discussion of theoretical foundations of the program from geography, urban planning, and psychology literature; and suggestions for improving program efficacy
